The Rab effector protein RabEPK, also known as Rab9 effector protein with kelch motifs (RABEPK/GAPEX-5), is a cytoplasmic protein involved in intracellular vesicle trafficking and endosomal sorting. It interacts with Rab9 GTPase, facilitating the recycling of mannose 6-phosphate receptors (M6PRs) from late endosomes to the trans-Golgi network. This process is critical for lysosomal enzyme transport and cellular homeostasis. Dysregulation of RabEPK has been linked to lysosomal storage disorders and neurodegenerative conditions.

RABEPK antibodies are immunological tools developed to study the protein's expression, localization, and functional interactions. They are widely used in techniques like Western blotting, immunofluorescence, and immunoprecipitation to investigate Rab9-mediated trafficking pathways. Commercial RABEPK antibodies are typically raised in rabbits or mice using recombinant protein fragments or synthetic peptides as immunogens. Validation often includes knockdown controls or colocalization studies with Rab9 and lysosomal markers.

Recent studies explore RABEPK's potential role in cancer, as altered vesicular trafficking contributes to tumor progression and drug resistance. Researchers also utilize these antibodies to characterize trafficking defects in cellular models of neurological diseases. Proper antibody validation remains crucial due to occasional cross-reactivity with structurally similar kelch domain-containing proteins.
