The ARPC5L (Actin-Related Protein 2/3 Complex Subunit 5-Like) antibody is a tool used to detect the ARPC5L protein, a component of the Actin-Related Protein 2/3 (ARP2/3) complex. This evolutionarily conserved complex plays a critical role in nucleating branched actin filament networks, which drive cellular processes like membrane protrusion, cell migration, and vesicular trafficking. ARPC5L, along with its paralog ARPC5. forms a heterodimer that stabilizes the ARP2/3 complex and enhances its actin-nucleating activity. While ARPC5 is ubiquitously expressed, ARPC5L exhibits tissue-specific expression, with higher levels in immune cells, testis, and spleen, suggesting specialized roles in immune regulation or spermatogenesis.

Studies using ARPC5L antibodies have helped elucidate its involvement in cytoskeletal dynamics and disease contexts. For instance, ARPC5L dysfunction has been linked to hematopoietic abnormalities, including T-cell deficiency and platelet anomalies, as well as certain cancers marked by aberrant cell motility. The antibody is commonly employed in techniques like Western blotting, immunofluorescence, and immunohistochemistry to assess ARPC5L expression levels, localization, and interactions. Its development has facilitated research into how ARP2/3-mediated actin remodeling contributes to physiological processes (e.g., immune cell activation) and pathological conditions (e.g., metastatic invasion). Validation of ARPC5L antibody specificity remains essential, given the high homology between ARPC5L and ARPC5.
