The GGA2 (Golgi-localized, γ-ear-containing, ARF-binding protein 2) antibody is a tool used to study the GGA2 protein, a member of the GGA family of clathrin adaptors involved in intracellular protein trafficking. GGAs regulate vesicle formation by sorting cargo between the Golgi apparatus, endosomes, and lysosomes. GGA2 contains three functional domains: a VHS domain for cargo recognition (e.g., mannose-6-phosphate receptors), a GAT domain that interacts with ARF GTPases and ubiquitin, and a hinge region connecting to a clathrin-binding γ-adaptin ear (GAE) domain. It plays a key role in maintaining cellular homeostasis by directing lysosomal enzyme transport and modulating receptor recycling/degradation.

GGA2 is implicated in neurodegenerative diseases, such as Alzheimer's, where dysregulation of trafficking pathways (e.g., APP processing) may contribute to pathology. Its expression and activity are also linked to cancer progression and viral infection mechanisms. The GGA2 antibody enables detection and localization of endogenous GGA2 in techniques like Western blotting, immunofluorescence, and immunohistochemistry. Studies using this antibody have elucidated GGA2's role in COP1 vesicle formation, AP-1 complex interplay, and stress-induced trafficking adaptations. Knockout models reveal compromised lysosomal function, underscoring its physiological significance. Researchers utilize this antibody to explore membrane trafficking dynamics and disease-related dysfunctions.
