DACT3 (Dapper Homolog 3. also known as DPR3) is a member of the Dapper (DACT) protein family, which modulates Wnt signaling and planar cell polarity pathways. Initially identified as a regulator of embryonic development, DACT3 interacts with key signaling components like β-catenin, Dishevelled (Dvl), and TGF-β pathways, influencing cell proliferation, differentiation, and tissue morphogenesis. Structurally, DACT3 contains conserved Dapper homology domains that mediate protein-protein interactions.

In cancer research, DACT3 has gained attention as a potential tumor suppressor. Epigenetic silencing of DACT3 via promoter hypermethylation is frequently observed in malignancies such as colorectal, liver, and gastric cancers. Loss of DACT3 expression correlates with Wnt/β-catenin pathway activation, promoting tumor progression and metastasis. Antibodies targeting DACT3 are essential tools for studying its expression patterns, subcellular localization, and functional roles in both physiological and pathological contexts.
