The DAOA (D-amino acid oxidase activator) gene, also known as G72. encodes a protein implicated in modulating D-amino acid oxidase (DAAO) activity, which regulates D-serine metabolism—a co-agonist of NMDA receptors critical for glutamatergic neurotransmission. First identified in the early 2000s through genetic linkage studies, DAOA gained attention for its potential association with psychiatric disorders, particularly schizophrenia and bipolar disorder. Variations in the DAOA gene have been linked to altered NMDA receptor function, contributing to the "glutamate hypothesis" of schizophrenia, which posits dysregulated glutamatergic signaling as a key pathogenic mechanism.

DAOA antibodies, specifically targeting the encoded protein, emerged as tools to study its expression, localization, and interaction with DAAO in neural tissues. Research suggests DAOA may act as a chaperone or stabilizer for DAAO, influencing D-serine levels and NMDA receptor activity. However, findings remain controversial, with inconsistent replication of its genetic association with psychiatric diseases. DAOA antibodies are also explored for diagnostic or therapeutic applications, though clinical utility is unproven. Current studies focus on clarifying DAOA's physiological role, its interaction with synaptic proteins, and potential as a biomarker or drug target in neuropsychiatric conditions. Challenges include understanding tissue-specific expression and resolving discrepancies between genetic and functional data.
