Death-associated protein kinase 1 (DAPK1) antibodies are immunological tools used to detect and study the DAPK1 protein, a calcium/calmodulin-dependent serine/threonine kinase involved in regulating apoptosis, autophagy, and cell survival. DAPK1 plays a tumor-suppressive role by promoting programmed cell death in response to cellular stress, DNA damage, or oncogenic signals. Its activity is often lost in cancers due to promoter hypermethylation, making it a potential biomarker for tumor progression and prognosis. Dysregulation of DAPK1 has also been linked to neurodegenerative diseases, such as Alzheimer’s disease, and ischemic brain injury.

DAPK1 antibodies are widely utilized in research to investigate its expression, localization, and post-translational modifications (e.g., phosphorylation at Ser308) across tissues and disease models. These antibodies enable techniques like Western blotting, immunohistochemistry (IHC), immunofluorescence (IF), and flow cytometry. Specific antibody clones may target distinct epitopes or functional domains, such as the N-terminal kinase domain or the C-terminal death domain. Recent studies explore DAPK1's interaction with cytoskeletal proteins, membrane receptors, and autophagy regulators, highlighting its therapeutic potential. Additionally, isoform-specific antibodies help differentiate DAPK1 from its splice variants (DAPK2. DAPK3) or phosphorylated forms, aiding mechanistic studies in cancer, neurology, and inflammation. Validation of antibody specificity remains critical to avoid cross-reactivity with homologous kinases.
