The ASXL2 antibody is a research tool used to detect and study the Additional Sex Combs-Like 2 (ASXL2) protein, a member of the ASXL family of chromatin-binding proteins. ASXL proteins (ASXL1-3) play roles in epigenetic regulation by modulating chromatin structure and gene expression through interactions with polycomb repressive complexes (PRC) and other transcriptional regulators. ASXL2. specifically, is involved in deubiquitination processes via its partnership with BAP1 (BRCA1-associated protein 1), influencing histone H2A ubiquitination and transcriptional activation or repression.

ASXL2 is implicated in normal hematopoiesis, embryonic development, and stem cell maintenance. Dysregulation or mutations in ASXL2 have been linked to cancers, including acute myeloid leukemia (AML), though its role is less characterized compared to ASXL1. ASXL2 antibodies are critical for investigating its expression patterns, subcellular localization, and functional interactions in both physiological and pathological contexts. These antibodies are commonly used in techniques like Western blotting, immunohistochemistry (IHC), and immunofluorescence (IF) to assess protein levels in cell lines or tissues. Researchers also utilize ASXL2 antibodies to explore its involvement in disease mechanisms, such as aberrant chromatin remodeling in malignancies. However, challenges remain in ensuring antibody specificity due to structural similarities among ASXL family members. Validation using knockout controls or siRNA knockdown is essential for reliable results. Ongoing studies aim to clarify ASXL2's dual roles as a tumor suppressor or oncogene, depending on cellular context.
