The VPREB3 antibody targets the VPREB3 protein, a component of the pre-B cell receptor (pre-BCR) complex critical during early B-cell development. VPREB3. encoded by the VPREB3 gene, belongs to the V-preB family, which includes surrogate light chain proteins that pair with the immunoglobulin heavy chain (μ chain) in precursor B cells. This interaction facilitates pre-BCR assembly, a key checkpoint for B-cell maturation, ensuring proper heavy chain expression and driving clonal expansion before light chain rearrangement.

VPREB3 is predominantly expressed in the bone marrow and lymphoid tissues, with restricted expression in pre-B cells. Its dysfunction has been linked to immunodeficiencies and B-cell malignancies. Antibodies against VPREB3 are essential tools in studying B-cell ontogeny, pre-BCR signaling, and diseases like acute lymphoblastic leukemia (ALL) or agammaglobulinemia. They enable detection of VPREB3 via techniques such as flow cytometry, Western blotting, and immunohistochemistry, aiding in diagnostics and mechanistic research. Commercial VPREB3 antibodies are typically validated for specificity using knockout cell lines or recombinant protein controls. Research using these antibodies has expanded insights into B-cell tolerance, leukemia biomarkers, and potential therapeutic targets.
