The ZNF274 antibody is a tool used to study the zinc finger protein 274 (ZNF274), a transcription factor belonging to the Krüppel-associated box (KRAB) domain-containing zinc finger protein (ZNF) family. ZNF274 is characterized by multiple C2H2-type zinc finger motifs, which facilitate sequence-specific DNA binding, and a KRAB domain that mediates interactions with co-repressors like KAP1 (TRIM28). This protein plays a role in transcriptional repression by recruiting chromatin-modifying complexes, such as the histone methyltransferase SETDB1. to establish heterochromatin and silence target genes, particularly retrotransposons and repetitive elements.

ZNF274 antibodies are widely employed in chromatin immunoprecipitation (ChIP) assays, immunofluorescence, and Western blotting to investigate its genomic localization, protein expression, and regulatory mechanisms. Studies using these antibodies have revealed ZNF274's involvement in maintaining genome stability and regulating imprinted gene clusters. For example, ZNF274 is enriched at telomeric regions and specific gene promoters, where it facilitates H3K9me3 deposition to suppress transcription.

The development and validation of ZNF274 antibodies often include specificity tests using knockout cell lines or competitive peptides. These reagents have been critical in elucidating ZNF274's role in diseases, including cancers and neurological disorders, where aberrant chromatin regulation is implicated.
