LYPLA2 (lysophospholipase 2) is a serine hydrolase enzyme belonging to the α/β-hydrolase fold family, primarily involved in lipid metabolism. It catalyzes the hydrolysis of lysophospholipids, including monoacylglycerols (MAGs) and fatty acyl esters, playing a critical role in regulating bioactive lipid signaling molecules. LYPLA2 is structurally related to LYPLA1. sharing 68% amino acid similarity, but exhibits distinct substrate preferences and cellular functions. Research highlights its importance in metabolic processes, such as energy homeostasis and fatty acid metabolism, with potential links to obesity, diabetes, and neurodegenerative disorders like Alzheimer’s disease.

LYPLA2 antibodies are essential tools for studying the enzyme's expression, localization, and activity in various tissues. These antibodies are widely used in techniques like Western blotting, immunohistochemistry, and immunofluorescence to investigate LYPLA2’s role in lipid signaling pathways. Recent studies have also explored LYPLA2’s interaction with endocannabinoid systems and its regulatory effects on autophagy. Commercially available LYPLA2 antibodies are typically validated for specificity using knockout cell lines or siRNA-mediated silencing. Understanding LYPLA2’s function through antibody-based research may provide insights into therapeutic strategies targeting metabolic or neurological diseases. However, challenges remain in distinguishing LYPLA2 from its homolog LYPLA1 due to their high sequence similarity, necessitating careful experimental design.
