RBFOX3. also known as NeuN (Neuronal Nuclei), is a neuron-specific RNA-binding protein encoded by the *RBFOX3* gene. It belongs to the RBFOX family of proteins involved in regulating alternative splicing and mRNA stability, particularly in neuronal tissues. RBFOX3 is widely recognized as a marker for postmitotic neurons, playing roles in neuronal maturation, synaptic function, and maintaining neuronal identity. Its expression is predominantly observed in the nucleus and cytoplasm of mature neurons across the central and peripheral nervous systems.

The RBFOX3 antibody is a critical tool in neuroscience research, commonly used to identify and label neuronal populations in both normal and pathological contexts. It detects a protein with a molecular weight of approximately 37-40 kDa, corresponding to splice variants of RBFOX3. This antibody has been validated in techniques such as immunohistochemistry (IHC), immunofluorescence (IF), and Western blotting (WB), aiding in studies of neurodevelopment, neurodegeneration (e.g., Alzheimer’s disease), and brain tumors. Its specificity for mature neurons makes it invaluable for distinguishing neuronal cells from glial or progenitor cells in tissue samples.

Discrepancies in staining patterns across species (e.g., humans, mice, rats) highlight the importance of antibody validation for experimental models. Recent studies also explore RBFOX3’s potential roles beyond a mere marker, including its involvement in RNA metabolism linked to neurological disorders.
