NOD2 (nucleotide-binding oligomerization domain-containing protein 2), also known as NLRC2. is a cytosolic pattern recognition receptor critical in innate immune responses. It detects bacterial peptidoglycan components, particularly muramyl dipeptide (MDP), and activates downstream signaling pathways like NF-κB and MAPK to trigger inflammatory responses. Structurally, NOD2 contains two N-terminal caspase recruitment domains (CARDs), a central NACHT domain for oligomerization, and C-terminal leucine-rich repeats (LRRs) responsible for ligand sensing.

NOD2 is highly expressed in immune cells (e.g., macrophages, dendritic cells) and epithelial cells, notably in the gastrointestinal tract. Genetic mutations in *NOD2* (e.g., frameshift, missense variants) are strongly linked to Crohn’s disease, Blau syndrome, and other autoinflammatory disorders. These mutations often impair MDP recognition or disrupt protein function, leading to dysregulated immune activation or defective bacterial clearance.

NOD2 antibodies are essential tools for studying its expression, localization, and signaling mechanisms. They enable detection of NOD2 in Western blotting, immunohistochemistry, and flow cytometry, aiding research on its role in inflammation, infection, and autoimmune diseases. Some antibodies target specific domains (e.g., LRR, CARD) to investigate functional or structural abnormalities caused by mutations. Additionally, NOD2 antibodies have potential diagnostic applications, helping identify aberrant expression in clinical samples, and may guide therapeutic strategies targeting NOD2-associated pathways in inflammatory bowel disease or cancer.
