背景信息

NOD2 (nucleotide-binding oligomerization domain-containing protein 2), also known as NLRC2. is a cytosolic pattern recognition receptor critical in innate immune responses. It detects bacterial peptidoglycan components, particularly muramyl dipeptide (MDP), and activates downstream signaling pathways like NF-κB and MAPK to trigger inflammatory responses. Structurally, NOD2 contains two N-terminal caspase recruitment domains (CARDs), a central NACHT domain for oligomerization, and C-terminal leucine-rich repeats (LRRs) responsible for ligand sensing.

NOD2 is highly expressed in immune cells (e.g., macrophages, dendritic cells) and epithelial cells, notably in the gastrointestinal tract. Genetic mutations in *NOD2* (e.g., frameshift, missense variants) are strongly linked to Crohn’s disease, Blau syndrome, and other autoinflammatory disorders.
