ATAD5 (ATPase family AAA domain-containing protein 5), also known as ELG1 in yeast, is a conserved protein critical for maintaining genomic stability during DNA replication. It belongs to the AAA+ ATPase family and functions as a key regulator of replication stress responses. ATAD5 forms a complex with the replication clamp loader (RFC) and plays a vital role in unloading proliferating cell nuclear antigen (PCNA) from chromatin after DNA synthesis. This process ensures proper recycling of PCNA, prevents replication fork stalling, and mitigates DNA damage accumulation.

Dysregulation of ATAD5 is linked to genomic instability and cancer. Studies show that ATAD5 acts as a tumor suppressor, with mutations or reduced expression observed in ovarian, endometrial, and colorectal cancers.
