ERAP1 (Endoplasmic Reticulum Aminopeptidase 1) is a key enzyme involved in antigen processing for major histocompatibility complex (MHC) class I presentation. It trims precursor peptides in the endoplasmic reticulum to their optimal length, enabling loading onto MHC class I molecules for immune surveillance by CD8+ T cells. Dysregulation of ERAP1 has been linked to autoimmune diseases, notably ankylosing spondylitis, psoriasis, and inflammatory bowel disease, as well as cancer progression. Genetic variations in ERAP1 are associated with altered enzymatic activity, influencing antigen repertoire selection and immune responses.
