DEFA5 (Defensin alpha 5) is a member of the α-defensin family, small cationic peptides critical in innate immunity. Primarily expressed in Paneth cells of the small intestine, DEFA5 exhibits broad-spectrum antimicrobial activity against bacteria, fungi, and enveloped viruses. It contributes to maintaining gut microbiota balance and defending against pathogens by disrupting microbial membranes. DEFA5 is also implicated in immune modulation, influencing cytokine production and immune cell recruitment.

Antibodies targeting DEFA5 are essential tools for studying its role in health and disease. They enable detection and quantification of DEFA5 in tissues and biofluids, aiding research on conditions like inflammatory bowel disease (IBD), colorectal cancer, and infections. In IBD, reduced DEFA5 expression correlates with dysbiosis and mucosal inflammation, highlighting its protective role. Commercially available DEFA5 antibodies are used in techniques such as ELISA, immunohistochemistry, and Western blot. However, challenges remain in ensuring specificity due to structural similarities among defensin family members. Recent studies explore DEFA5 as a potential biomarker or therapeutic target, emphasizing its dual function in microbial defense and immune regulation. Ongoing research aims to clarify its mechanisms in tissue-specific immunity and its translational applications.
