MANF (Mesencephalic Astrocyte-derived Neurotrophic Factor) is a secreted protein initially identified for its neuroprotective effects, particularly on dopaminergic neurons, which are critical in Parkinson’s disease. Unlike classical neurotrophic factors (e.g., GDNF or BDNF), MANF lacks a traditional receptor-binding mechanism and is structurally classified as a member of the conserved CDNF/MANF protein family. It plays a key role in endoplasmic reticulum (ER) stress response, interacting with unfolded proteins and modulating the unfolded protein response (UPR) to promote cell survival under stress conditions.

MANF antibodies are tools widely used in research to detect and quantify MANF expression in tissues or cells, aiding studies on its role in neurodegenerative diseases, diabetes, inflammation, and cancer. These antibodies enable techniques like Western blotting, immunohistochemistry, and ELISA, helping to map MANF distribution in the brain, pancreas, and other organs. Notably, MANF’s dual role in neuroprotection and ER stress regulation has spurred interest in therapeutic applications, with antibodies sometimes used to block or trace MANF in experimental models. Challenges remain in understanding its precise mechanisms and receptor interactions, but MANF antibodies remain pivotal in unraveling its potential as a biomarker or therapeutic target for conditions like Parkinson’s, stroke, and metabolic disorders.
