The CBFA2T2 antibody targets the CBFA2T2 protein (also known as MTG16), a member of the ETO family of nuclear transcriptional corepressors. CBFA2T2 plays critical roles in hematopoietic development, differentiation, and epigenetic regulation by forming complexes with chromatin-modifying enzymes (e.g., histone deacetylases) and transcription factors. It is implicated in transcriptional repression through interactions with DNA-binding proteins like RUNX1.

CBFA2T2 gained prominence due to its involvement in chromosomal translocations in acute myeloid leukemia (AML), particularly the t(16;21)(q24;q22) translocation that fuses CBFA2T2 with RUNX1. disrupting normal hematopoiesis. This fusion protein interferes with myeloid differentiation, contributing to leukemogenesis.
