ACTL6B (Actin-like 6B), also known as BAF53B, is a subunit of the neuron-specific BAF (Brg1/Brm-associated factor) chromatin remodeling complex, which regulates gene expression by modifying chromatin structure. As a member of the actin-related protein (Arp) family, ACTL6B shares structural homology with β-actin but lacks filament-forming activity. It plays a critical role in neural development, synaptic plasticity, and cognitive functions by facilitating the assembly and targeting of the BAF complex to specific genomic loci. Dysregulation of ACTL6B has been linked to neurodevelopmental disorders, including intellectual disability and autism spectrum disorders (ASD), as well as certain cancers.

Antibodies against ACTL6B are essential tools for studying its expression, localization, and interactions in cellular contexts. They are widely used in techniques such as Western blotting, immunofluorescence, and chromatin immunoprecipitation (ChIP) to investigate its role in chromatin remodeling and transcriptional regulation. Given its tissue-specific _expression (primarily in the brain), ACTL6B antibodies help distinguish neural BAF complexes from ubiquitously expressed BAF complexes containing the paralog ACTL6A (BAF53A). Research using these antibodies has advanced understanding of epigenetic mechanisms in neurodevelopment and disease, highlighting ACTL6B as a potential therapeutic target for neurological conditions.
