The GYS2 antibody targets glycogen synthase 2 (GYS2), a key enzyme in glycogen biosynthesis. GYS2. primarily expressed in the liver, catalyzes the transfer of glucose monomers from UDP-glucose to growing glycogen chains via α-1.4-glycosidic bonds, a critical step in hepatic glycogenesis. Unlike its isoform GYS1 (muscle-specific), GYS2 activity is regulated by allosteric effectors and hormonal signaling, particularly insulin, which promotes glycogen storage postprandially. Dysregulation of GYS2 is linked to glycogen storage diseases (GSDs), such as GSD 0a, characterized by fasting hypoglycemia and hyperketonemia due to impaired glycogen synthesis. Mutations in the GYS2 gene are associated with this rare autosomal recessive disorder, underscoring the enzyme’s role in glucose homeostasis.

GYS2 antibodies are essential tools for studying glycogen metabolism, enabling detection of protein expression, localization, and post-translational modifications (e.g., phosphorylation at Ser7/Ser10) that modulate enzyme activity. They are widely used in Western blotting, immunohistochemistry, and immunofluorescence to investigate hepatic glycogen dynamics in metabolic disorders, including diabetes and non-alcoholic fatty liver disease (NAFLD). Additionally, these antibodies aid in exploring GYS2’s potential role in hepatocellular carcinoma, where altered glycogen metabolism may influence tumor progression. Commercial GYS2 antibodies are typically validated for specificity in human and model organisms (e.g., mice, rats), supporting translational research aimed at therapeutic targeting of glycogen-related pathways.
