The TET2 (Ten-Eleven Translocation 2) antibody is a crucial tool in epigenetic research, targeting the TET2 protein, a member of the TET enzyme family. TET2 catalyzes the oxidation of 5-methylcytosine (5mC) to 5-hydroxymethylcytosine (5hmC) in DNA, a key step in active DNA demethylation. This process regulates gene expression, cellular differentiation, and genomic stability. TET2 mutations are frequently observed in hematologic malignancies, such as acute myeloid leukemia (AML), myelodysplastic syndromes (MDS), and lymphoma, linking its dysfunction to tumorigenesis and clonal hematopoiesis.

TET2 antibodies are widely used in techniques like Western blotting, immunohistochemistry (IHC), and immunofluorescence (IF) to detect TET2 expression levels, localization, and post-translational modifications in tissues or cell lines. They help elucidate TET2's role in developmental biology, cancer, and immune regulation. Researchers also utilize these antibodies to study correlations between TET2 loss/mutation and disease progression or therapeutic resistance.

Commercial TET2 antibodies are typically raised against specific epitopes, such as the C-terminal catalytic domain, and validated using knockout controls to ensure specificity.
