The POLR2K antibody targets a subunit of RNA polymerase II (Pol II), the essential enzyme responsible for transcribing protein-coding genes in eukaryotes. POLR2K, also known as RPB10. is a highly conserved, small subunit (∼8.2 kDa) that forms part of the core structure of Pol II. Unlike larger catalytic subunits, POLR2K plays a structural role in stabilizing the polymerase complex and facilitating interactions with transcription regulatory factors. It is encoded by the POLR2K gene located on human chromosome 8q21.11.

Research using POLR2K antibodies primarily focuses on understanding transcriptional regulation mechanisms. These antibodies are employed in techniques like Western blotting, chromatin immunoprecipitation (ChIP), and immunofluorescence to detect POLR2K expression, localize it within the nucleus, and study its association with Pol II during transcription initiation/elongation. Dysregulation of POLR2K has been implicated in cellular stress responses and diseases linked to transcription errors, including certain cancers and neurodegenerative disorders. For example, altered POLR2K expression patterns have been observed in hepatocellular carcinoma and glioblastoma.

Commercial POLR2K antibodies are typically raised against synthetic peptides corresponding to conserved regions of the human protein. Validations include specificity testing using knockout cell lines and cross-reactivity assessments across species. Researchers should optimize experimental conditions due to the subunit’s low molecular weight and potential cross-reactivity with other polymerase subunits. These reagents serve as critical tools for investigating fundamental gene expression processes and their pathological disruptions.
