VPS33B (Vacuolar Protein Sorting 33 Homolog B) is part of the HOPS (Homotypic Fusion and Protein Sorting) and CORVET (Class C Core Vacuole/Endosome Transport) complexes, which regulate membrane fusion events in endolysosomal and autophagy-related processes. This protein plays a critical role in maintaining lysosomal function, endosomal sorting, and intracellular vesicle transport, particularly in coordinating interactions between SNARE proteins and Rab GTPases during membrane fusion.

Mutations in the VPS33B gene are linked to arthrogryposis-renal dysfunction-cholestasis (ARC) syndrome, a rare autosomal recessive disorder characterized by multisystemic defects.
