The TGM4 antibody targets transglutaminase 4 (TGM4), a calcium-dependent enzyme belonging to the transglutaminase family. TGM4 is primarily expressed in the prostate gland and plays a role in cross-linking seminal fluid proteins, contributing to semen coagulation and sperm motility. Unlike other transglutaminases (e.g., TGM2 in celiac disease), TGM4 has limited tissue distribution, with high specificity to reproductive tissues. Research on TGM4 antibodies has gained interest due to their potential applications in studying prostate physiology, male infertility, and prostate cancer. In prostate cancer, aberrant TGM4 expression has been linked to tumor progression and metastasis, making it a candidate biomarker or therapeutic target.

Commercially available TGM4 antibodies are typically monoclonal or polyclonal, developed using recombinant TGM4 protein fragments. They are used in techniques like Western blotting, immunohistochemistry, and ELISA to detect TGM4 expression in tissue samples or biological fluids. Challenges in TGM4 antibody development include ensuring specificity, as cross-reactivity with other transglutaminases can occur due to structural similarities. Recent studies also explore autoantibodies against TGM4 in autoimmune contexts, though their clinical relevance remains under investigation. Overall, TGM4 antibodies serve as critical tools for unraveling the enzyme's biological roles and its implications in reproductive and oncological research.
