The TPSB2 antibody targets tryptase beta-2 (TPSB2), a serine protease primarily expressed in mast cells. Tryptases, including TPSB2. are stored in mast cell granules and released upon activation, playing key roles in allergic responses, inflammation, and tissue remodeling. TPSB2 is notable for its stability and enzymatic activity, cleaving substrates like fibrinogen, neuropeptides, and cytokines. Its dysregulation is implicated in mast cell-associated disorders, such as asthma, anaphylaxis, urticaria, and mastocytosis, as well as tumor microenvironments where mast cells influence cancer progression.

The TPSB2 antibody is widely used in research to detect and quantify TPSB2 protein levels, aiding studies on mast cell activation, disease mechanisms, and biomarker identification. It is applied in techniques like immunohistochemistry, ELISA, and Western blotting. Commercially available antibodies vary in specificity, with some distinguishing TPSB2 from its isoform TPSAB1 (alpha/beta tryptase). Recent studies also explore TPSB2 as a therapeutic target, with inhibitors under investigation for inflammatory and fibrotic diseases.

In clinical diagnostics, TPSB2 antibodies help confirm systemic mastocytosis by detecting elevated serum tryptase. However, cross-reactivity with other tryptases requires careful validation. Overall, the TPSB2 antibody serves as a critical tool for advancing both basic research and translational applications in mast cell biology.
