The TATA-box binding protein-associated factor 12 (TAF12) is a critical subunit of the TFIID complex, a general transcription factor essential for RNA polymerase II-mediated gene transcription. As part of the TFIID complex, TAF12 interacts with TBP (TATA-box binding protein) and other TAFs to facilitate promoter recognition, chromatin remodeling, and assembly of the pre-initiation complex. TAF12 contains a conserved histone-fold domain that enables dimerization with TAF6. forming a structural scaffold for TFIID. Beyond TFIID, TAF12 is also found in other transcriptional coactivator complexes, such as STAGA and TFTC, highlighting its versatile role in regulating gene expression.

TAF12 antibodies are widely used in research to investigate transcriptional mechanisms, protein-protein interactions, and chromatin dynamics. They are essential tools in techniques like chromatin immunoprecipitation (ChIP), co-immunoprecipitation (Co-IP), and Western blotting to study TAF12’s localization, binding partners, and expression levels. Dysregulation of TAF12 has been implicated in cancers and developmental disorders due to its role in controlling cell cycle and differentiation-related genes. For example, TAF12 overexpression or mutations may disrupt transcriptional fidelity, contributing to oncogenesis. Researchers also utilize TAF12 antibodies to explore its crosstalk with histone acetyltransferases and other epigenetic modifiers, providing insights into disease pathways and potential therapeutic targets. Its molecular weight (~20-25 kDa) varies slightly across species due to post-translational modifications.
