The REC8 antibody is a crucial tool in studying meiosis and chromosomal dynamics. REC8. a meiosis-specific cohesin component, plays a pivotal role in sister chromatid cohesion, chromosome pairing, and segregation during gametogenesis. It belongs to the kleisin family of cohesin proteins but is distinct from mitotic cohesins (e.g., RAD21/SCC1), as it is preferentially expressed in germ cells. During meiosis, REC8 forms a proteinaceous scaffold that maintains chromatid cohesion until anaphase II, ensuring proper chromosome segregation and genetic diversity through recombination. Its regulated proteolytic cleavage by separase enables stepwise chromosome separation.

REC8 antibodies are widely used to visualize meiotic chromosome architecture in model organisms (e.g., mice, yeast, plants) and human germ cells. They help identify meiotic defects linked to infertility, miscarriage, or chromosomal disorders like aneuploidy. In research, these antibodies facilitate studies on synaptonemal complex formation, crossover regulation, and the unique cohesion persistence required for biphasic meiosis. Commercial REC8 antibodies are typically developed against conserved epitopes in N-terminal regions, with validation in Western blot, immunofluorescence, and chromosome spread staining. Recent applications extend to cancer biology, as some tumors show ectopic REC8 expression potentially linked to genomic instability.
