The PSMC1 (Proteasome 26S Subunit, ATPase 1) antibody targets a critical component of the 26S proteasome, a large multiprotein complex responsible for regulated protein degradation in eukaryotic cells. PSMC1. also known as Rpt2. is part of the 19S regulatory particle that binds to the 20S core to form the 26S proteasome. It functions as an ATPase, providing energy for substrate unfolding and translocation into the proteolytic core. PSMC1 plays a key role in maintaining protein homeostasis, regulating processes like cell cycle progression, DNA repair, and stress response by degrading ubiquitinated proteins.

Antibodies against PSMC1 are widely used to study proteasome composition, dynamics, and dysfunction in diseases. They enable detection of PSMC1 expression levels via techniques like Western blotting, immunofluorescence, and immunohistochemistry. Research applications include investigating proteasome activity in cancer (where proteasomes are often hyperactive), neurodegenerative disorders (linked to protein aggregation), and immune responses. Altered PSMC1 expression has been associated with drug resistance in chemotherapy and the efficacy of proteasome inhibitors like bortezomib. These antibodies also help explore evolutionary conservation of the ubiquitin-proteasome system across species. Proper validation ensures specificity for distinguishing PSMC1 from other AAA-ATPase family members in the 19S complex.
