PARP11 (Poly(ADP-ribose) polymerase 11) is a member of the PARP protein family, which plays diverse roles in DNA repair, chromatin remodeling, and cellular stress responses. Unlike well-characterized PARPs like PARP1 and PARP2. PARP11 is less studied but has unique functional features. It contains a conserved PARP catalytic domain and a distinct regulatory domain, enabling its involvement in non-canonical pathways. Research indicates PARP11 localizes to the nuclear envelope and interacts with components of the nuclear pore complex, suggesting a role in nucleocytoplasmic transport or mitotic progression. It has also been linked to interferon signaling and immune regulation, with potential implications in viral defense and autoimmune diseases.

PARP11 antibodies are critical tools for investigating these functions. They enable detection of PARP11 expression via Western blotting, immunofluorescence, or immunohistochemistry, and facilitate studies on its subcellular localization and protein interactions. Specificity validation (e.g., using PARP11-knockout controls) is essential due to structural similarities among PARP family members. Recent studies highlight PARP11's possible oncogenic role in certain cancers, driving interest in its therapeutic targeting. However, its mechanistic contributions remain unclear, underscoring the need for reliable antibodies to advance functional and clinical research. Commercial PARP11 antibodies are typically raised against unique epitopes within its N-terminal region, ensuring minimal cross-reactivity. Ongoing work aims to clarify PARP11's biological significance and explore its potential as a biomarker or drug target.
