The IPO4 antibody targets Importin-4 (IPO4), a member of the importin β family of nuclear transport receptors. These proteins facilitate the transport of cargo molecules, such as proteins or RNAs, across the nuclear membrane via the nuclear pore complex. IPO4 specifically mediates the nuclear import of cargoes bearing a classical nuclear localization signal (NLS), often partnering with importin-α adaptors to recognize NLS-containing substrates. It plays roles in diverse cellular processes, including transcriptional regulation, cell cycle progression, and signal transduction.

IPO4 antibodies are widely used in research to study nuclear-cytoplasmic transport mechanisms, protein localization, and disease pathways linked to transport dysregulation, such as cancer or viral infections. They are employed in techniques like Western blotting, immunoprecipitation, and immunofluorescence to detect IPO4 expression levels, subcellular distribution, or interactions with cargo proteins. Structural studies indicate that IPO4 shares conserved features with other importins, including a helical HEAT-repeat domain for cargo binding and RanGTPase interaction.

Notably, IPO4 has been implicated in the nuclear import of specific regulators, such as the steroid hormone receptors and SMAD proteins, highlighting its functional versatility. Antibodies against IPO4 help elucidate its tissue-specific expression, post-translational modifications, and potential as a therapeutic target. Most commercial IPO4 antibodies are raised against human or murine epitopes, with validation in common experimental models.
