The 15-hydroxyprostaglandin dehydrogenase (HPGD) antibody is a key tool for studying the enzyme responsible for prostaglandin catabolism. HPGD, also known as 15-PGDH, catalyzes the oxidation of prostaglandins, including PGE2 and PGF2α, into biologically inactive 15-keto metabolites, thereby regulating their signaling. This NAD+-dependent enzyme is expressed in various tissues, such as the lung, liver, placenta, and kidney, and plays a critical role in maintaining prostaglandin homeostasis.

HPGD antibodies are widely used in techniques like Western blotting (WB), immunohistochemistry (IHC), and ELISA to detect protein expression levels, tissue localization, and activity modulation. Researchers employ these antibodies to explore HPGD's involvement in pathological processes, including inflammation, fibrosis, and cancer. For instance, reduced HPGD expression has been linked to tumor progression in colorectal and bladder cancers due to elevated prostaglandin signaling.

Commercial HPGD antibodies are typically raised in rabbits or mice against synthetic peptides or recombinant protein fragments. Validation includes specificity checks via knockout controls and functional assays. Their applications extend to studying NSAID mechanisms, tissue regeneration, and metabolic disorders. Proper handling with protease inhibitors is recommended for tissue lysate-based experiments.
