The HOMER2 antibody is a tool used to study the HOMER2 protein, a member of the HOMER family of scaffolding proteins critical for organizing signaling complexes at postsynaptic densities in neurons. HOMER proteins (HOMER1. HOMER2. HOMER3) exist in two forms: constitutively expressed short isoforms and activity-dependent long isoforms. HOMER2. encoded by the *HOMER2* gene, interacts with glutamatergic receptors, including metabotropic glutamate receptors (mGluRs) and inositol trisphosphate receptors (IP3Rs), to regulate intracellular calcium signaling, synaptic plasticity, and neurotransmitter release. Its structure includes an N-terminal EVH1 domain for binding proline-rich motifs in target proteins and a C-terminal coiled-coil domain enabling dimerization.

HOMER2 antibodies are widely employed in neuroscience research to investigate synaptic function, neuropsychiatric disorders, addiction mechanisms, and neurodegenerative diseases. For example, HOMER2 is implicated in alcohol and drug addiction due to its role in reward-related signaling pathways. Antibodies against HOMER2 enable detection of its expression and localization via techniques like Western blotting, immunohistochemistry, and immunoprecipitation. Researchers also use these antibodies to study HOMER2's interaction with proteins like mGluR5 or Shank, shedding light on synaptic dysfunction in conditions such as autism spectrum disorders and Alzheimer’s disease. Specificity validation (e.g., knockout controls) is essential, as HOMER isoforms share structural homology.
