HOMER1 is a member of the HOMER family of scaffolding proteins, which play critical roles in organizing synaptic signaling complexes at the postsynaptic density (PSD) of neurons. Discovered in the late 1990s, HOMER1 is encoded by the *HOMER1* gene and exists as multiple splice variants, including the immediate-early gene product HOMER1a and constitutively expressed isoforms like HOMER1b/c. These proteins contain an N-terminal Enabled/VASP homology 1 (EVH1) domain that binds to proline-rich motifs in target proteins, such as metabotropic glutamate receptors (mGluRs), and a C-terminal coiled-coil domain enabling self-oligomerization. This structure allows HOMER1 to link membrane receptors to intracellular calcium stores, ion channels, and downstream signaling effectors, modulating synaptic plasticity, calcium signaling, and receptor trafficking.

HOMER1 antibodies are essential tools for studying the expression, localization, and function of HOMER1 proteins in neurological and psychiatric research. They are widely used in techniques like Western blotting, immunohistochemistry, and immunofluorescence to investigate synaptic alterations in conditions such as Alzheimer’s disease, epilepsy, and schizophrenia. Dysregulation of HOMER1 expression has been linked to synaptic dysfunction, cognitive deficits, and behavioral abnormalities, making these antibodies valuable for exploring disease mechanisms. Commercially available HOMER1 antibodies are typically validated for specificity against target isoforms, though challenges remain in distinguishing splice variants due to shared epitopes.
