ALDH1A1. ALDH1A2. and ALDH1A3 belong to the aldehyde dehydrogenase 1A subfamily, which catalyzes the oxidation of retinaldehyde to retinoic acid (RA), a critical signaling molecule in development, differentiation, and homeostasis. These cytosolic enzymes share structural homology but exhibit distinct tissue expression and functional roles. ALDH1A1 is widely expressed in adult tissues, including the liver, brain, and stem cells, and is recognized as a marker for cancer stem cells in various malignancies. ALDH1A2. predominantly expressed during embryogenesis, is essential for RA synthesis in neural crest cell development and heart morphogenesis. ALDH1A3 shows restricted expression in the brain, retina, and certain glands, playing roles in neurogenesis and ocular development.

Antibodies targeting these isoforms are vital tools for studying their tissue-specific expression, RA signaling pathways, and disease associations. ALDH1A1 antibodies are frequently used in cancer research to identify stem-like tumor populations linked to therapy resistance. ALDH1A2 antibodies help investigate developmental defects, such as congenital heart anomalies, while ALDH1A3 antibodies are employed in studies on neurodevelopmental disorders or retinal degeneration. However, cross-reactivity remains a challenge due to high sequence similarity (~70% homology) among isoforms. Validation via knockout controls or isoform-specific peptides is critical to ensure antibody specificity. These antibodies support diverse applications, including Western blotting, immunohistochemistry, and flow cytometry, advancing research into developmental biology, cancer mechanisms, and regenerative medicine.
