Poly(ADP-ribose) polymerase 2 (PARP2) is a member of the PARP enzyme family, which plays critical roles in DNA repair, genome stability, and cellular stress responses. PARP2 shares structural and functional similarities with PARP1. including a conserved catalytic domain responsible for poly(ADP-ribosyl)ation (PARylation), a post-translational modification involved in DNA damage signaling. However, PARP2 has distinct N-terminal domains that influence its substrate specificity and interaction partners. It participates in base excision repair (BER), chromatin remodeling, and transcriptional regulation, with emerging roles in inflammation, metabolism, and cell death pathways.
