The HIC2 (Hypermethylated in Cancer 2) antibody is a tool used to study the HIC2 protein, encoded by the HIC2 gene located on human chromosome 22q11.21. HIC2 functions as a transcriptional regulator belonging to the BTB/POZ zinc finger protein family, implicated in diverse biological processes, including cell proliferation, differentiation, and apoptosis. Initially identified as a tumor suppressor, HIC2 is frequently epigenetically silenced via hypermethylation in various cancers, such as colorectal and breast cancer, correlating with poor prognosis. Beyond oncology, HIC2 plays critical roles in cardiovascular development, particularly in heart morphogenesis and outflow tract septation, as evidenced by studies in mouse models.

The HIC2 antibody is widely utilized in research to detect and quantify HIC2 protein expression via techniques like Western blotting, immunohistochemistry (IHC), and immunofluorescence (IF). It aids in exploring HIC2's interaction networks, subcellular localization (primarily nuclear), and regulatory mechanisms. Researchers also employ this antibody to investigate HIC2's role in DNA damage response and its cross-talk with signaling pathways like p53 and Wnt. Commercial HIC2 antibodies are typically validated for specificity using knockout controls or siRNA-mediated knockdown. Despite its utility, challenges remain in standardizing detection protocols across tissue types due to variable expression levels. Ongoing studies aim to clarify HIC2's dual roles in tumor suppression and developmental biology, highlighting its potential as a therapeutic target or diagnostic marker.
