ERAP2 (Endoplasmic Reticulum Aminopeptidase 2) is a key enzyme involved in antigen processing for major histocompatibility complex class I (MHC-I) presentation. It trims precursor peptides in the endoplasmic reticulum to optimize their length and binding affinity for MHC-I molecules, enabling immune recognition by CD8+ T cells. ERAP2 operates in tandem with its homolog ERAP1. though their substrate preferences and trimming mechanisms differ. Genetic polymorphisms in ERAP2 have been linked to autoimmune and inflammatory diseases, including ankylosing spondylitis, Behçet's disease, and psoriasis, highlighting its role in immune regulation. Dysregulation of ERAP2 may also influence viral infection outcomes and cancer immunity by altering antigenic peptide repertoires.

Antibodies targeting ERAP2 are critical tools for studying its expression, localization, and functional interactions. They enable detection of ERAP2 in cellular models, assessment of enzymatic activity modulation, and exploration of disease-associated genetic variants. Specific ERAP2 antibodies (e.g., monoclonal clones like 4E7 or polyclonal variants) are validated for techniques including Western blotting, immunohistochemistry, and immunofluorescence. Recent research explores ERAP2 inhibition as a therapeutic strategy for autoimmune disorders or cancer immunotherapy, driving demand for high-specificity antibodies to evaluate drug efficacy.
