The NSUN4 antibody is a research tool designed to detect and study the NSUN4 protein, a member of the NOL1/NOP2/Sun (NSUN) family of RNA methyltransferases. NSUN4 is primarily localized in mitochondria and plays a critical role in mitochondrial ribosome biogenesis by catalyzing the methylation of mitochondrial ribosomal RNA (rRNA), specifically the m5C modification at position 911 of 12S rRNA. This modification is essential for proper assembly of the mitochondrial ribosome and subsequent translation of mitochondrial DNA-encoded proteins, which are vital for oxidative phosphorylation and cellular energy production.

NSUN4 interacts with mitochondrial transcription factor MTERF4. forming a complex necessary for stabilizing rRNA structure and ensuring efficient ribosome assembly. Dysregulation of NSUN4 has been linked to mitochondrial dysfunction, which is implicated in metabolic disorders, neurodegenerative diseases, and cancer. For instance, NSUN4 deficiency disrupts mitochondrial protein synthesis, leading to impaired respiratory chain activity and compromised cellular energy metabolism.

The NSUN4 antibody is widely used in techniques like Western blotting, immunofluorescence, and immunohistochemistry to investigate NSUN4 expression, localization, and its role in mitochondrial biology.
