The PSMB10 antibody targets the proteasome subunit beta 10 (PSMB10), a key component of the immunoproteasome. PSMB10. also known as β2i or LMP10. is a catalytic subunit within the β-ring of the 20S immunoproteasome, a specialized proteasome variant induced by cytokines like interferon-gamma (IFN-γ). Unlike constitutive proteasomes, immunoproteasomes are primarily expressed in immune cells and play a critical role in generating antigenic peptides for major histocompatibility complex (MHC) class I presentation. PSMB10 enhances the cleavage specificity of proteasomes, favoring hydrophobic and basic residues, which optimizes antigen processing for immune surveillance.

PSMB10 antibodies are widely used to study immunoproteasome dynamics in autoimmune diseases, neurodegenerative disorders, and cancer. Dysregulation of PSMB10 has been linked to conditions such as rheumatoid arthritis, Alzheimer’s disease, and tumor immune evasion. Researchers employ these antibodies in techniques like Western blotting, immunohistochemistry (IHC), and flow cytometry to assess protein expression, localization, and immunoproteasome activity.

Commercial PSMB10 antibodies are typically raised in rabbits or mice, validated for specificity against recombinant human PSMB10. Studies using these antibodies have elucidated the immunoproteasome's role in inflammation, protein homeostasis, and therapeutic responses, making them essential tools in immunology and biomedical research.
