The OTX1 antibody targets the Orthodenticle homeobox 1 (OTX1) protein, a transcription factor belonging to the homeodomain-containing protein family. OTX1 plays critical roles in embryonic development, particularly in the central nervous system (CNS) and sensory organ formation. During neurodevelopment, it regulates the patterning and differentiation of the forebrain and midbrain. Studies also link OTX1 to retinal development and craniofacial morphogenesis. Beyond development, OTX1 is implicated in pathological processes, including cancer. Overexpression of OTX1 has been observed in tumors such as glioblastoma, medulloblastoma, and melanoma, where it may drive proliferation, invasion, and therapy resistance by modulating pathways like Wnt/β-catenin or PI3K/AKT.

OTX1 antibodies are widely used in research to study its expression, localization, and function via techniques like Western blotting (WB), immunohistochemistry (IHC), immunofluorescence (IF), and flow cytometry. These antibodies are typically raised in rabbits or mice, with specificity validated through knockout controls or siRNA knockdown. Commercially available clones vary in host species, clonality (monoclonal/polyclonal), and epitope recognition. Researchers rely on OTX1 antibodies to explore its dual role in developmental biology and disease, particularly its potential as a diagnostic marker or therapeutic target in cancers. However, challenges remain in ensuring antibody specificity due to homology with OTX2. a closely related family member. Ongoing studies aim to clarify OTX1’s context-dependent functions and its interplay with other regulatory proteins.
