Interleukin-18 binding protein (IL18BP) is a soluble decoy receptor that specifically neutralizes the pro-inflammatory cytokine IL-18 by preventing its interaction with cell surface receptors. IL-18. a member of the IL-1 cytokine family, plays a key role in inflammatory and immune responses, particularly by inducing interferon-gamma (IFN-γ) production and promoting Th1-mediated immunity. Dysregulated IL-18 signaling is linked to autoimmune disorders (e.g., rheumatoid arthritis, lupus), inflammatory diseases, and certain cancers. IL18BP, naturally produced by the body, acts as a critical regulatory mechanism to control excessive IL-18 activity.

IL18BP antibodies are tools developed to detect, quantify, or modulate IL18BP in research and therapeutic contexts. In research, these antibodies enable the study of IL18BP expression patterns, interactions with IL-18. and its role in disease models. Therapeutically, recombinant IL18BP or IL18BP-targeting antibodies are explored to either enhance IL-18 neutralization (e.g., in cytokine storm syndromes) or block IL18BP to boost IL-18-driven anti-tumor immunity in cancer immunotherapy. Monoclonal antibodies against IL18BP are designed for high specificity, with some in preclinical development for inflammatory and oncological indications. Challenges include optimizing binding affinity and understanding tissue-specific IL18BP dynamics. Overall, IL18BP antibodies represent a promising avenue for modulating IL-18 pathways in immune-related diseases.
