The SLC2A4 antibody targets the solute carrier family 2 member 4 (SLC2A4), also known as glucose transporter type 4 (GLUT4), a critical insulin-regulated transporter responsible for facilitating glucose uptake into adipose and muscle tissues. GLUT4 is stored in intracellular vesicles under basal conditions and translocates to the cell membrane upon insulin stimulation, a process central to maintaining glucose homeostasis. Dysregulation of GLUT4 expression or trafficking is closely associated with insulin resistance, type 2 diabetes, and metabolic syndrome.

SLC2A4 antibodies are widely used in research to detect GLUT4 protein levels, localization, and dynamics in tissues or cell models. They enable techniques like Western blotting, immunohistochemistry, and immunofluorescence to study GLUT4 membrane translocation, expression changes in metabolic disorders, or responses to therapeutic interventions. These antibodies are often raised in hosts like rabbits or mice, targeting specific epitopes within GLUT4’s cytoplasmic, extracellular, or C-terminal domains.

Validation of SLC2A4 antibodies is essential, as variations in isoform specificity, cross-reactivity, or sensitivity can affect experimental outcomes. Knockout controls or siRNA-mediated silencing are recommended to confirm antibody specificity. Research utilizing SLC2A4 antibodies has advanced understanding of insulin signaling pathways, adipose-muscle crosstalk in metabolism, and potential drug targets for diabetes. However, challenges remain in standardizing GLUT4 quantification due to its low basal membrane presence and tissue-specific expression patterns.
