MYOD1 (myoblast determination protein 1) is a pivotal transcription factor encoded by the MYOD1 gene, belonging to the family of myogenic regulatory factors (MRFs). It plays a central role in skeletal muscle differentiation by activating muscle-specific gene expression and reprogramming non-muscle cells into myoblasts. MYOD1 antibodies are essential tools in biomedical research, primarily used to detect and quantify MYOD1 protein levels in various experimental models. These antibodies are widely applied in studies of myogenesis, muscle regeneration, and muscle-related diseases, such as rhabdomyosarcoma, where MYOD1 expression is often dysregulated.

In diagnostics, MYOD1 antibodies aid in identifying tumors with skeletal muscle differentiation, as MYOD1 is a lineage-specific marker. Commercially available antibodies typically target epitopes within the N-terminal transactivation domain or the C-terminal basic helix-loop-helix (bHLH) domain, critical for DNA binding and dimerization. Validation methods include Western blotting, immunohistochemistry (IHC), and immunofluorescence (IF), often using cell lines like C2C12 or tissues with known MYOD1 expression.

Recent studies also explore MYOD1's role in regenerative medicine and cellular reprogramming. However, antibody specificity remains a consideration, as cross-reactivity with other MRFs (e.g., Myogenin) may occur. Researchers prioritize monoclonal antibodies for consistency or polyclonal ones for broader epitope recognition. Quality control, including knockout validation, ensures reliable results. Overall, MYOD1 antibodies remain indispensable for unraveling muscle biology and advancing therapeutic strategies for muscular disorders.
