The EZH1 antibody is a key tool for studying the function of Enhancer of Zeste Homolog 1 (EZH1), a catalytic subunit of the Polycomb Repressive Complex 2 (PRC2). EZH1 shares homology with EZH2. both functioning as histone methyltransferases responsible for trimethylating lysine 27 on histone H3 (H3K27me3), a hallmark of transcriptionally repressive chromatin. However, EZH1 and EZH2 exhibit distinct biological roles. While EZH2 dominates PRC2 activity in proliferating cells, EZH1 is more prominent in post-mitotic or differentiated cells, acting as a modulator of gene silencing in tissues with low cell turnover, such as skeletal muscle, neurons, and hematopoietic stem cells. This functional divergence arises from differences in their enzymatic kinetics and PRC2 complex-binding preferences.

EZH1 antibodies are widely used to investigate its role in epigenetic regulation, developmental processes, and diseases, particularly cancer. Studies reveal that EZH1 can compensate for EZH2 loss in certain contexts, contributing to tumor progression or suppression depending on cellular conditions. Researchers employ these antibodies in techniques like chromatin immunoprecipitation (ChIP), Western blotting, and immunofluorescence to map EZH1’s genomic targets, quantify expression, or assess its subcellular localization. Validation of antibody specificity is critical due to structural similarities between EZH1 and EZH2. often requiring knockout controls or isoform-specific validation assays. Recent interest in EZH1 stems from its emerging therapeutic relevance, as dual EZH1/EZH2 inhibitors are being explored for cancer treatment.
