LILRA1 (leukocyte immunoglobulin-like receptor A1), also known as CD85I, is a member of the LILR family of immunoregulatory receptors primarily expressed on myeloid cells, including monocytes, macrophages, and dendritic cells. It belongs to the activating subgroup of LILRs, characterized by a charged residue in the transmembrane domain that facilitates association with signaling adaptors like FcRγ. LILRA1 interacts with classical and non-classical MHC class I molecules (e.g., HLA-A, HLA-G) and other ligands, modulating immune responses through dual functions: promoting pro-inflammatory cytokine production or exerting inhibitory effects depending on cellular context and ligand engagement.

Antibodies targeting LILRA1 are valuable tools for investigating its role in immune regulation, particularly in autoimmune diseases, infections, and cancer. Studies show LILRA1 is involved in autoimmune conditions like rheumatoid arthritis and systemic lupus erythematosus, where aberrant signaling may disrupt immune tolerance. In viral infections (e.g., HIV), LILRA1 interacts with viral proteins to influence immune evasion. In cancer, tumor-associated macrophages often upregulate LILRA1. potentially contributing to immunosuppressive microenvironments. Research-grade anti-LILRA1 antibodies enable detection of receptor expression, functional blocking/activation assays, and exploration of therapeutic targeting strategies. Recent interest focuses on developing monoclonal antibodies to modulate LILRA1 activity, aiming to restore immune balance in diseases or enhance antitumor immunity. However, functional complexity and ligand diversity necessitate careful characterization of antibody specificity and downstream effects.
