The MUC5B antibody is a critical tool for studying the MUC5B mucin, a high-molecular-weight, heavily glycosylated protein encoded by the *MUC5B* gene. As a major component of mucus, MUC5B plays essential roles in forming protective gel-like layers on epithelial surfaces, particularly in the respiratory tract, salivary glands, and cervix. It contributes to lubrication, pathogen trapping, and immune defense. Dysregulation of MUC5B expression is linked to pathologies such as chronic obstructive pulmonary disease (COPD), idiopathic pulmonary fibrosis (IPF), and certain cancers. Notably, a promoter polymorphism in *MUC5B* (rs35705950) is strongly associated with IPF risk, driving research interest in its pathogenic mechanisms.

MUC5B antibodies are widely used in techniques like immunohistochemistry, Western blotting, and ELISA to detect MUC5B’s localization, expression levels, and glycosylation patterns. These antibodies help distinguish MUC5B from other mucins (e.g., MUC5AC) and elucidate its unique roles in health and disease. In respiratory research, they aid in exploring mucus hypersecretion in asthma or bronchitis, while in cancer studies, they investigate MUC5B’s dual role as a tumor suppressor or promoter, depending on context. Commercial MUC5B antibodies are typically raised against specific protein regions or glycosylated epitopes, requiring validation for target specificity.
