SPI1 (also known as PU.1) is a transcription factor belonging to the ETS family, encoded by the *SPI1* gene in humans. It plays a pivotal role in hematopoiesis, particularly in the development and differentiation of myeloid and lymphoid lineages. SPI1 regulates the expression of genes critical for immune cell function, including those involved in B-cell maturation, macrophage differentiation, and granulocyte development. It binds to purine-rich DNA sequences in promoter/enhancer regions, modulating transcriptional activity through interactions with cofactors like C/EBPα and GATA proteins. Dysregulation of SPI1 is linked to hematologic malignancies, such as acute myeloid leukemia (AML), where mutations or altered expression disrupt normal cellular differentiation.
