TOP2B (Topoisomerase II Beta) is a nuclear enzyme belonging to the type II topoisomerase family, which plays a critical role in DNA replication, transcription, and chromosome segregation by transiently breaking and rejoining double-stranded DNA to relieve torsional stress. Unlike its isoform TOP2A, which is predominantly expressed in proliferating cells and linked to cell cycle regulation, TOP2B is ubiquitously present in both dividing and non-dividing cells, particularly in neurons and post-mitotic tissues. This isoform is implicated in neurodevelopment, synaptic plasticity, and transcriptional regulation, with emerging roles in neurological disorders such as Alzheimer’s disease and ALS.

Antibodies targeting TOP2B are essential tools for studying its expression, localization, and function in diverse biological contexts. They are widely used in techniques like Western blotting, immunohistochemistry, and immunofluorescence to investigate TOP2B's involvement in DNA repair mechanisms, chromatin remodeling, and responses to genotoxic stress. Additionally, TOP2B antibodies have clinical relevance in cancer research, as aberrant TOP2B expression or activity is associated with certain malignancies, including neuroblastoma and treatment-resistant tumors. Notably, TOP2B is a secondary target for some chemotherapeutic agents (e.g., etoposide), though its inhibition is often linked to off-target toxicity rather than therapeutic effects. Research utilizing these antibodies continues to clarify TOP2B's dual roles in maintaining genomic stability and contributing to disease pathogenesis, bridging molecular biology with therapeutic development.
