The PAF1 (RNA polymerase II-associated factor 1) antibody is a crucial tool for studying the PAF1 complex, a multifunctional protein assembly conserved from yeast to humans. This complex, comprising subunits like PAF1. LEO1. CTR9. CDC73. and RTF1. plays pivotal roles in transcriptional regulation, histone modification, and chromatin remodeling. It interacts directly with RNA polymerase II (Pol II) to facilitate transcription elongation, mRNA processing, and termination. The PAF1 subunit, in particular, is essential for recruiting histone-modifying enzymes, such as Set1/COMPASS (for H3K4 methylation) and Bre1 (for H2B ubiquitination), thereby influencing epigenetic landscapes.

PAF1 antibodies are widely used in techniques like immunoprecipitation, ChIP-seq, and immunofluorescence to investigate the complex's localization, interactions, and regulatory mechanisms. Dysregulation of the PAF1 complex is implicated in cancers, developmental disorders, and stem cell maintenance defects. For example, PAF1 overexpression correlates with tumor progression in certain cancers, while mutations in associated subunits (e.g., CDC73) link to hyperparathyroidism-jaw tumor syndrome. Researchers also employ PAF1 antibodies to explore its role in cell cycle control, DNA repair, and pluripotency.
