RHOH (Ras homolog family member H) is a small GTPase belonging to the Rho subfamily, primarily expressed in hematopoietic cells. Unlike canonical Rho GTPases, RHOH lacks intrinsic GTPase activity due to amino acid substitutions in conserved catalytic domains, rendering it constitutively GTP-bound. It plays critical roles in immune cell signaling, particularly in T-cell development, activation, and thymocyte selection. RHOH regulates cytoskeletal dynamics, cell migration, and antigen receptor signaling by interacting with downstream effectors like WASP and ZAP-70.

Antibodies targeting RHOH are essential tools for studying its expression, localization, and function in immune regulation. They enable detection via techniques such as Western blot, immunofluorescence, and flow cytometry. Research using RHOH antibodies has linked its dysregulation to hematological malignancies (e.g., lymphoma, leukemia) and immune disorders. For instance, RHOH mutations or aberrant expression disrupt normal T-cell receptor signaling, contributing to immunodeficiency or autoimmunity. Commercial RHOH antibodies are typically validated for specificity using knockout cell lines or siRNA-mediated silencing. Recent studies also explore RHOH's role in cancer metastasis and its potential as a therapeutic target, emphasizing the continued relevance of high-quality antibodies in dissecting RHOH-related pathways.
