The ALG2 antibody is designed to target the ALG2 (alpha-1.3-mannosyltransferase) protein, a key enzyme involved in N-linked glycosylation, a critical post-translational modification process. ALG2 catalyzes the addition of the second mannose residue to the lipid-linked oligosaccharide (LLO) precursor in the endoplasmic reticulum (ER), a step essential for proper glycoprotein synthesis. Mutations in the ALG2 gene are associated with congenital disorders of glycosylation (CDG), specifically CDG-Ii, a rare autosomal recessive disease characterized by multisystemic abnormalities, including neurological deficits, developmental delays, and impaired immune function.

ALG2 antibodies are primarily utilized in research to study glycosylation mechanisms, detect ALG2 expression in cellular or tissue samples, and investigate pathological pathways in CDG-Ii. They are also valuable tools for diagnosing CDG subtypes via genetic and protein analysis. Commercially available ALG2 antibodies are typically developed in rabbit or mouse hosts, validated for applications such as Western blotting, immunofluorescence, or immunohistochemistry.

Understanding ALG2’s role and its antibody’s specificity aids in elucidating glycosylation-related diseases and developing targeted therapeutic strategies. However, antibody validation remains crucial due to potential cross-reactivity with homologous glycosyltransferases, emphasizing the need for precise epitope mapping and functional assays in experimental workflows.
